Product Description:
Proteinase K is a non-specific serine protease derived from the fungus Engyodontium album (formerly Tritirachium album). This product is free of DNase and RNase contamination and can efficiently digest a wide range of proteins.
The enzymatic activity of this product is ≥30 Units/mg. One unit (U) is defined as the amount of enzyme that liberates 1 μmol of tyrosine per minute from casein substrate at 37 °C and pH 7.5. Proteinase K exhibits broad tolerance, with an effective working pH range of 4.0-12.5 and an optimal reaction pH of 7.5-8.0. Its optimal reaction temperature is 65 °C; however, high temperature tends to cause autodegradation of the enzyme. A reaction temperature of 50-55 °C is recommended for routine experiments.
The commonly used working concentration is 0.05-1 mg/mL, which can be adjusted flexibly according to system pH, temperature and components such as SDS and urea. Common reagents including EDTA, Triton X-100, Tween 20, Sarkosyl and guanidine hydrochloride have minimal impact on its enzymatic activity. It features excellent system compatibility and is suitable for digestion of various complex samples.
Applications:
l Catalyzes hydrolysis of peptide and ester bonds; enables peptide synthesis in non-aqueous solvents.
l Elimination of endogenous nucleases in nucleic acid preparation, genomic DNA extraction.
l Pre-treatment of tissue sections for in situ hybridization.
