DMEM (Dulbecco's Modified Eagle Medium) is developed based on the MEM (Minimum Essential Medium). Compared with the MEM medium, the amino acid content is doubled, the vitamin content is quadrupled, and non-essential amino acids, trace iron ions, and sodium pyruvate are additionally supplemented. It is a widely used basic culture medium that can support the growth of various types of mammalian cells. The original DMEM formula contains a relatively low concentration (1g/L) of glucose and sodium pyruvate. However, there are currently multiple DMEM formulations available, including those with higher glucose concentrations and those with or without sodium pyruvate. This product contains various components necessary for cell culture, such as amino acids, vitamins, and inorganic salts, but does not contain proteins, lipids, or growth factors. Therefore, this product must be used in combination with serum or serum-free additives, and usually 10%–20% fetal bovine serum (FBS) is added.
DMEM is available in three formulations based on glucose concentration: High-glucose DMEM (4.5 g/L), Low-glucose DMEM (1.0 g/L), and glucose-free DMEM.
